Peptidoglycan Biosynthesis

D-alanine is a crucial component of the peptidoglycan layer, which provides structural integrity to bacterial cell walls.
It is incorporated into:

Stem peptides (as part of the tetrapeptide chain: L-alanine → D-glutamate → L-lysine/meso-diaminopimelate → D-alanine-D-alanine).
Cross-links between peptidoglycan strands (via transpeptidation).

Formation of D-Ala-D-Ala Dipeptide

Two D-alanine molecules are joined by the enzyme D-alanine-D-alanine ligase (Ddl) to form D-alanyl-D-alanine, a key substrate for peptidoglycan synthesis.
This dipeptide is added to UDP-MurNAc-tripeptide to form UDP-MurNAc-pentapeptide, a peptidoglycan precursor.

Target for Antibiotics

Since D-alanine is unique to bacteria (not found in humans), enzymes involved in its metabolism are targets for antibiotics:

Vancomycin binds to the D-alanyl-D-alanine terminus, blocking transpeptidation.
D-cycloserine inhibits alanine racemase (converts L-alanine → D-alanine) and D-alanine-D-alanine ligase, disrupting peptidoglycan synthesis.
